Tidus is forcing out laughter cause he knows he is approaching the fact that he is going to have to murder his father who disappeared 10 years ago. He hates his father because his mother was enthralled with him and when he disappeared she gave up on life.

Yuna is forcing out her laughter cause she will die if she succeeds in killing Sin which happens to be Tidus father.

I never made it far in that game, but I did know the scene was forced laughter...just never knew the full details. :3
