I don't know if an entire playable job will created out of the aftermath of the Blasphemies, at least no more so the previous expansions new jobs depended on their respective locations.

Also considering the soul stone mechanic of jobs, there will likely be some pre-established in-universe history to the job (Machinists would technically be the "youngest" of all jobs with soul stones, right?)

Yes, Machinist is currently known to be the youngest Job founded in the Source due to only recently being created in Ishgard by the current Machinist Guildmaster Stephanivien. It is to a point the MCH job crystal is literally a clean slate and WoL is just putting his/her experience into the Job Crystal instead of the Crystal giving WoL knowledge.

However, this also means other regions may also create new Jobs as well and may have establish new organizations for these new Jobs that we are unaware about due to having no knowledge of those regions currently.

The Demon Hunter job establishment may not be so far as well since we know very little about the unknown regions such as the New World. It is mostly a matter of establishing lore about what the new job was before they create Job Crystals to preserve the knowledge of their combat skills. For Demon Hunter Job case, they can go down the path where they started out as normal monster hunters but when the final days hit and people started turning into Blasphemies these Hunters used their skills to fight off the Blasphemies and save people. In the aftermath of such events, they may have established their new organization dedicated to hunting down Blasphemies but in these new regions they may call Blasphemies "Demons" instead thus leading to the name Demon Hunters for the Demon Hunter Job.

However, this is only one path they may take if they consider introducing Demon Hunter Job and lore. I don't expect such a job to actually happen but there is a lot of questions running through my mind now about the aftermath for the Final Days for regions that do not have the information about Season 1 events due to how little contact they have with Eorzea and regions that do make contact with Eorzea.
