For the most part it isn't the end of the world if you miss your positional. But the damage increase is drastically significant comparing MNK to the other melee jobs.

That depends on the positional.
Missing a True Strike or Twin Snakes for example isn't a big deal at all, while missing either Fang and Claw or Wheeling Thrust is a big deal since it's both the extra damage + no Raiden Thrust.
On Monk the one you need to pay extra attention to is Bootshine but you also have Riddle of Earth on top of the usual positional ignore.

Edit: I realize you're probably talking about just ignoring them altogether and not just missing one lol.
It didn't even occur to me because people not even putting in the effort at all and just standing in one place is so nonsensical to me that it didn't even cross my mind until I read what you were responding to.

But it's not just the extra potency either, on SAM for example that Kenki adds up too and you get less OGCD's.
I've never done this because it's just dumb, but I am sure if people have the parse addon they can test it themselves and see the difference.
I am pretty sure it's significant even without testing it myself.

I'd probably say that NIN is the one with the least significant positionals.
