It really has been years since I've been through some of these questlines and I was wondering, of all the jobs currently in-game, which are the ones in which "YOU are THE [insert job here] and no one else (or very few else)"?

I wouldn't say any job is unique to the WoL per see, but there are some degrees of scarcity.
- WHM is a job that is common place but usually limited to the Padjali, ergo a non Padjal WHM would be considered unqiue. BLM is rare for the whole fobidden arts sctick
- RDM and MNK were more aboundant but their numbers dwindled to a few ar msot after the fall of Ala Mhigo
- I'm certain that jobs like DRG, PLD and especially MCH are commonplace, if limited to a few locales
Depends. Some Jobs are stated as being unique or lost when you first unlock them, but as the story goes along others become inspired by your actions and accomplishments and take up the Job themselves (usually because the NPC you gain the Job from starts teaching others). MNK, SCH and SMN are examples of this.
Some Jobs are taught by secretive organisations or orders that usually are closed off to outsiders, but have started teaching their arts to adventurers who 'catch their eye' and have distinguished themselves (AST, RDM, SAM, PLD, DRK and NIN are examples).
Some Jobs are literally lost or restricted and so you are literally the only 'authorized' practioner of it (WHM is an example of this, as the Job was made forbidden after Amdapor's misuse of the Job during the War of the Magi where they created life, but was entrusted to the padjal of Gridania for safe keeping by the elementals. BLM is also an example in rather similar circumstances).
And some Jobs have been made literally out of whole cloth then and there (MCH and BLU were both brand new Jobs which you're basically experimenting with on the creator's behalf - this is why their Soul Crystals are listed in the Equipment menu as being empty of past accomplishments).
DRG is an interesting example all of it's own though as although there are 'normal' DRGs which look identical to your version (effectively Ishgard's version of LNC but with extra dragonslaying skills), the WoL technically is in a league of their own, as they are ordained as an 'Azure DRG', a role that is meant to be unique (Estinien is meant to be the Azure DRG, but the Eye of Nidhogg annointed the player as one too, something that had never happened before). Given Estinien destroyed the Eyes of Nidhogg for good as part of the story, this set of circumstances will never be repeated, thus making the WoL (and their DRG Soul Crystal) unique.
DRK is also rather unique in although the Job is stated as being an order of outcast knights that lurk in the shadows despensing justice to those considered above the law in Ishgard, the player gains the Job in a rather unusual set of circumstances that is likely to not occur again. So although there are other DRKs cannonically, a WoL DRK is unique (not helped by that whole dark side becoming independent of your psyche thing).
Either way, each Job varies between ones which others canonically can use and others that are literally unique, that you are the only user of that Job.

None of the jobs are truly unique to the WoL. Every single job has one or more other members with the job (often our Rival, if not teacher also has the job). At most you could say in some cases the WoL is a the first modern or Eorzean member of a job.
Currently the only job that fits the description of the WoL being the only current practitioner is Scholar, though that may change in the future if Setoto manages to attune to Lily again.

DRK because there's only 1 other.
RDM because there's only 1 other, +1 in training and Alisaie is a RDM, but not a Crimson Duelists so I don't know how that counts. Alisaie doesn't wear the red.
WTM because there's only 2 others I think? It's suppose to be Padjal only.
DRG, while there are many Dragoons, there's only 2 other "Azure Dragoons" (one of whom, I don't think is officially a "Azure Dragoon")
GNB, it's unclear how rare this job is. The one who teaches you thinks he's the last of his kind, only to find others during the job quests.
Alisaie was trained by the same person we did, she doesnt need to wear red either she can still be a crimson duelist but most importantly shes one of the Main Characters, having gear that shows off her job isnt required.DRK because there's only 1 other.

Padjals are fairly common as well, theres the girl from SB WHM Quests/the Conjurer trainer/Kan-e-Senna/the 2 WHM Padjals that train us early/another one at Quarymill too
At lvl 80[?] i think the GNB trainer mentions hes been training adventurers who got interested in the class as well so yeah doubt hes alone.
Now that I think on it, the WoL isn't a Crimson Duelist so that wouldn't even count, even if I'm wearing the red. The Crimson Duelists are a defunct organization.Alisaie was trained by the same person we did, she doesnt need to wear red either she can still be a crimson duelist but most importantly shes one of the Main Characters, having gear that shows off her job isnt required.
