Yes the research on Echo has been going on for a long time now in Garlemald.
While originally they killed Echo users because of belief that they were no different from Primals, the 1.0 Warriors of Light actions for being able to kill Primals changed this view and now the Garlean Empire see the Echo as a possible solution to their "Primal threat" thus they began experimenting on how to replicate the Echo and learning more about.
This has been going for some time since 2.0 as there have been hints Garlean Empire have been reasearching the Echo. It is only now we see the fruit of their labor with experimenting on Echo users in Garlemald.
We have no way of knowing what the next xpac is, until it is announced. I'm sure it will be easier to guess, the closer we get to 5.0, but as of now it looks like it could go one or more of many different ways. 1. A Bukyo Expansion, where we deal with Hingashis attempt at reform. The caste system is apparently a huge problem, leading to civil unrest. 2. Garlemald. Obviously that Garlemald Embassy has to be set up for later. We're also at war with them. 3. Thavnair. Out of all the other known nations, we get a Thavnair embassy, plus it's on the map. It's very possible. 4. More of the east opens up, allowing us to travel to Naxcia, The Burn, and more unreleased places. 5. Sharlayan opens up for some reason. 6. The Roe origin land of Aerslant. 7. Meracydia.
Regula is the mask just assume the obvious since we know of only one person close to Varis. Anyways on the scientist he's the second boss in Ala Mhigo.
I recall them saying Garlemald's story wouldn't be done until 6.0 at the earliest, so I could see 5.0 mostly being a continuation of the war against them, pressing onwards into the regions within outer Othard and southern Ilsabard, capitalizing on the civil unrest stirred up by Doma and Ala Mhigo's liberation.

Ya for 4.0 we are shown the worst case of Garlean Control areas but as certain NPCs said in 4.0, through side quests and main story, Garlean Control areas vary on who is the one in control of the area. For some areas they get Good Garleans as their rulers which in return these areas are treated better and given reason to find their life better under Garlean rule compared to Doma and Ala Mhigo which they only got beatings and empty promises.
