I am having some difficulty getting past the second part of the fight for this quest when I am ask to give Alisaie my aether.
I cast all my aether on her and the duty still fails.
Am I misunderstanding what I am supposed to do here?

You have to do it more than once. Each scion will take turns over and over. You have to watch when your turn comes up each time.
Also, I've noticed a lot of people don't read the NPC text and realize that on the last phase the Ranger will focus target Alisae and kill her unless you focus on stopping the Ranger. Make sure this isn't happening to you.
There is a meter thing that tells you how much aether you've given to Alisaie; you have to get the meter up to 100% before your opponent's does or else it's game over (pay attention to know when it's your turn to give aether).
Moreover, like Velox said, you have to stop The Ranger of Darkness from killing Alisaie or else her casting will fail and it's a wipe.

I just did this quest, took 5 attempts my issue was I couldn't even see the clickable item.... I have never been so frustrated in this game than last night trying to complete that Duty
That was my main issue as well, and more so in that the hint given in the dialogue was actually misleading how it kept telling me to give it to Alisaie. Worse, the Blade of Light's target has it's on screen text in white not green, meaning it blended in with the aetheric energy effect swirling around, and I didn't realize this til it was far too late (I kept trying to click on Alisaie instead and wondered why nothing was happening).
The other issue with that fight is 'Evil!Derplander' (the WAR) decided he simply hates you and will tend to focus all his attention on you, not helped by the first part of the fight he has an Invincible effect which makes him no sell anything you throw at him. And in the second stage he has the annoying tendency to interrupt you when trying to target said Blade of Light (worse if the PLD has decided to tag team with him), meaning you have no choice but to beat him down just to have some breathing space.
But really the only issue was the target needed to click on was too easily obscured. Other than that it was fairly straightforward (not to mention satisfying ;)).
My brother has the same problem now. He wiped over 10 times now because there is no "blade of light".
We both watched on the screen for the whole fight and ran around but we cant' find this stupid blade. Where is it?
I can't remember having problems to find it while i was doing it myself. It was just in front of her. But now there is nothing.
He is close to uninstalling the game because he is so frustrated right now. And i can fully understand him.
Make sure the nameplates for objects isn't set to hidden.
I just did it again on another char and I made sure I was in front of her and clicking on the bottom half of her body area to give aether. That is the worst pos quest I ever did. The devs need to check themselves.
Horrible quest, horrible battle design... hard to see spot to click and terrible instructions. I hate cheap quests that don't give you a chance to figure it out and give you a crappy mechanic that tries to make you fail.
THIS ONE OF THE MOST F****** WORST QUESTS I HAVE EVER PLAYED IN THIS GAME, MAKES ME WANA RAGE QUIT, WHAT A PILE OF CRAP
Now I have calmed down, click on the floor in front of her feet to give the either, very hard to or not possible to try click the sword its self. It took 4 attempts and no quest should be this frustrating as its neither challenging or fun.
You should just have to click her and not have to try and find the sweet spot on where to click!
Are there other ways you can select targets on PC? I'm on PS4 and you can use the left/right D-pad buttons to precisely target the correct object.
Not liking it mechanically is one thing. Not liking it narratively is another.
It was a great questline, biggest flaw was it was so short.
This is ridiculous. I've been trying to do this damn quest for an hour now because I have to keep searching the internet for solutions after every faill. It's been 4 years since the first post and they haven't been able to fix this? The developers are really fkn incompetent.

It's actually a really easy quest. You just have to pay attention and put a little effort into your attempts. The aether draw point is the only non-NPC you can target in the fight. Just tab on through.
I have just dug this post up because I am on my 7th try and still can't do this terrible quest. Even when it's supposedly my turn and someone isn't hitting me I can't activate the sword no matter where I click. I think the dev's need to fix this quest as it seems hit and miss and with it being a MSQ it should work properly.
Theres nothing to fix because, again, it isnt broken. It will work the first time you do it correctly provided you dont die during the fighting. If you have object nameplates hidden, turn them on for this instance. If you need to tab through targets or use controller target cycling, do it. The aether blade youre powering up is forming a couple feet in front of Alissaie.
I just did it a week ago on my latest alt. Had no problems completing the duty.
It's not broken, it seems to be more a misunderstanding by some players about what is supposed to happen.
You're only going to be able to click when you see "Blade of Light" appear. If the name does not show, you won't be able to channel your aether. Alisaie will call for more aether a few seconds before the name will appear.
Remember that it is a channel, not a simple click and run. It takes about 5 seconds to complete the channel. Interrupt it before it's done and you have to start over.
If you can't get it targeted and interact with a mouse click, use your Confirm keybind (default is NUM0 on a standard keyboard, handy trick for interacting with any object/NPC in an area crowded with players or enemy NPCs). You need to make sure you don't have anything else targeted, you are in range and directly facing it, and that there are no Scions standing between you and the Blade of Light. Press your Confirm key once to target the Blade then a second time to start channeling.
Hope this helps if you haven't already completed the duty.
this shouldve been "fixed" long ago. ppl defending the design of this quest, for what?? all the complaints are completely valid. the "fix" is make the interactable object more visible. is that hard?
The interactable object is the front the aether field around Alisaie. It is the only one, and it works like any other interactable object in any other place. Quite literally just go stand next to the Elezen girl who is surrounded by white-blue aether that takes up half of the entire battlefield. Sheathe your weapon and tab or press X until you interact with the Blade of Light, and let the cast bar fill completely. Easy. It couldn't be anymore visible. And the NPCs go to near the same spot when they do it, too.
The hang up is players not paying attention, sorry not sorry.
Stupid quest, doing damage, no Aether, doing heal no Aether. Seems to be random with the Aether but not enough to finish.
Sadly this is a campaign quest. So far all quests in the campaign had been more or less nicely done, but this one is garbage.
Worked now after several trys, did it with heal and damage. But still just random appear of the Aether at Alisea, so i cant give any help to other players who have the same problem. Please rename this quest to random success.
I know this effects a tiny fraction of the population, but I am a gamer with low vision and I think I will never succeed at this quest.

I can think of something that might help you but I need to run through the quest in NG+ to test it first. I'll come back to edit this post with the method if it works.
It did work so hopefully it will work for you as well.
When the Blade of Light appears to you the first time in the fight (about 20 seconds after the second fight starts), take a moment to mark it with a Sign (found in the Party menu). You can assign Signs to an empty hotbar space if you have one to make them easier to access during the fight. You might want to play with the Signs on a NPC or other object before you enter the duty to see which is most visible to you. Remember that there's going to be a lot of blue-white glowing around Alisaie so you'd want to pick one that stands out in that.
Once the channel is completed, the Blade of Light and Sign will disappear but the Sign will still be on the Blade of Light when it reappears. Hopefully that will make it easier for you to find and target the Blade the second and third times in the fight.
Try to stay away from Alisaie except when it's time to channel. Focus on killing the Devout and Ranger every time they're resurrected. Don't attack the other 3 before the first channel. After the first channel, kill the Knight after the Devout and Ranger are dead. Don't attack the Warrior and Magus. After the second channel, kill the Warrior (and possibly the Knight) once the Devout and Ranger are dead. If the Ranger begins to target Alisaie (you'll see a gold line appear between them), make it a priority. Dodge AoEs as needed.
It's not broken, but just has mechanics that are very difficult. The strategy for this duty works as others have mentioned, so unless you're trying to do it in gear that is too low ilevel, even if you're failing you should be able to pass it eventually, due to the Echo boost effect kicking in that should superpower your stats, and does stack if you're failing repeatedly and retrying it.
Also, are you keeping the duty set as 'Normal' difficulty after failing it? There is nothing to gain keeping it on 'normal' if you're having trouble with this duty unless you're deliberately looking for a challenge. Set it to 'Very Easy' and it should help you also a great deal.
Yeah, every person before you said the same thing and was proven wrong. So... im going to stick to the assumption that you arent doing it right as you have provided no details to the contrary. And look at that! One post before yours in this very thread someone listed not only a kill order for the NPCs, but a tip to make the most important part of the fight(channeling the aether blade) easier. We all agree that this fight could have been designed better, but it is perfectly clearable as is. And who knows. Theyve been updating bits and pieces of the MSQ as they go. This patch might be looked at again via the 6.3 patch in 4ish months. It COULD be improved. But again, it doesnt need to be fixed. Cause it aint broke.
I don't get why people are being so condescending about this quest.
I know there's supposed to be a Blade of Light in front of Alisaie. I know that it has no physical body, and I have to click the name directly. I know that I need to defeat anybody who's aggro'd on me or her, or we won't be able to focus on channeling. And yet, even when I do that, there are times when the Blade of Light just...isn't there. How do I make it reappear so that I can continue interacting with it? It's so frustrating having the NPCs bark orders for me to do something I can't do because the item has not respawned yet. "GET UP ON THE HYDRA'S BACK, GET UP ON THE HYDRA'S BACK,' except the entire hydra has temporarily despawned.
Like, do people realize it's possible to pay attention and adore this game and still have valid criticisms of encounter design? I'm not used to seeing the community behave so dismissively and throw some imagined 'fault' onto new players like they're doing in this thread.
I know I am a necromancer for clearing palace solo, but you're a necromancer in your own right with posts being from 2+ years ago. I don't remember ever failing this quest nor am I going to go back and read a thread from over TWO YEARS AGO to understand why people are stuck.
If you're failing, try again or lower the difficulty.
As I recall the Blade of Light indeed isn't always on the screen, but only becomes available when it's actually scripted into the battle as needed, based on how the battle progresses and how you fill up the gauge (you can tell when it's now available when Alisaie says something like "lend me your aether" or something along those lines. That's when you have to stop what you're doing and focus on the Blade of Light - of course with Darklander and his flunkies snapping at your heels, this can be easier said than done.
And yes, it's a very difficult battle, but it's not impossible - I failed three or four times myself until I finally realized what I was doing wrong (didn't actually notice the Blade of Light when it became available, and then couldn't click on it in time). Once I managed to get a handle on that mechanic, I eventually passed it.
So don't despair, the fight is hard, but use the advice that is given, and the knowledge that the Blade isn't always on the screen but will become available at scripted points in the fight and that is when you have to click on it to feed it your aether to boost the gauge, then you'll soon emerge victorious (battered and bruised though).
