Aren't you, english speakers (frenchy here) using "thou" as you / your's ? Meaning he would be saying somethin like "what are your words master Matoya ?"
I would go for "Matoya" being a title or a family name given by Master Matoya to Yshto'la for some reasons

My Theory:
I think the scene with Uriangier and Y’Shtola/Matoya is the closest to us in terms of timeframe.
Uriangier is ‘sent back’ from wherever the scions have been ‘called’, he acts as a messenger, and reveals the coming calamity. We seek out Matoya’s help, who decides to fight alongside us ‘until Y’shtola returns’. But she can’t do that in her frail old body, so she chooses her star pupil who she knows would agree to being used as a vessel in this manner under dire circumstances.
That's more or less the meaning - basically "what do you say [about this], Master Matoya?"
(Or normally we'd probably ask "what do you think?")
The exact line is "what sayest thou" - it's an older form of English that uses different endings on verbs, like in German. (I'm a native English speaker but learned German at school.)
And yes, "thou" means "you".
