I think Samurai could be tank, Red Mage as healer, and Blue Mage as ranged DPS.

I think RDM would be more of a BRD/MCH type of Job (MP Refresh/TP Regain), but as a melee DPS.
I really hope we don't get XIs version of a Red mage, but since a classic RDM wouldn't really fit in the holy trinity I guess that's probably the closest thing we'll get. You never know tho
IIRC Yoshida said in the past that they wanted either dark knight or samurai to be a tank, and they chose dark knight. That would presumably mean samurai will be melee DPS, especially since there hasn't been a new strength DPS class added since 1.0.
I think he's also said that they won't ever add 3 jobs at once again, so it'll probably be either RDM or BLM and not both.
Red Mage is basically confirmed. Not only the Red Mage, Blue Mage debate at the door, but...

I thought Doomtrain was just a minion they were going to release. The final boss of a few Final Fantasies... I'm hoping Hades or Ozma!
Eh. Hades and Ozma were optional bosses in Final Fantasy IX exclusively -- which isn't to say they won't ever appear. Hades has been a summon in prior games, but never a boss. Ozma never appeared outside of IX, but could be interesting Allagan technology. I just don't think Ozma would be a priority, though.
Phantom Train seems much more likely because the Phantom Train already exists in Final Fantasy XIV. Its name was mentioned a handful of times that it was carrying magitek weaponry past Ul'dah and into Northern Thanalan. It's very possible that because the Garlean soldiers have lost the lives of so many of their comrades, that they'd be desperate enough to pray for resources and supplies to fight, and with the amount of Aether in the region it's quite possible that those prayers could summon Doomtrain/Phantom Train.
On a somewhat yet totally unrelated note,
I would love for some of the official Final Fantasy XIV preview videos to end with a song or sound effect that tells of what's to come if there isn't video ready for it. I think all the fanboys and fangirls would go crazy to hear something like the sound of the Phantom Train at the end of a video, or any sound effect or song that hints at what's to come. The Internet would tear it apart in a split second and create a hype... train?

Hades is also the final boss of FFXI's Seekers of Adoulin. So he has been a boss in FFIX, FFXI and a summon in FFVII. Phantom Train is a boss in FFVI and has made cameo appearances in FFI and FFIV: The After Years as an optional boss (but I still count that as only one boss as it's a cameo. The Hades bosses in FFIX and FFXI are completely different). Doomtrain (Glasya Labolas) has been a summon in only FFVIII.
