In both of these quests, you are supposed to throw the key item at the glowy spots. You are not supposed to right-click the glowy spots.

To throw the key item at the spot, use the key item by left clicking the icon between the quest name and the exclamation mark in your duty list. For example, for the Rock of Rancor quest, the icon that you should click looks like a wooden box. It is small, but it is there. After clicking it, your mouse cursor turns into a targeting reticle (similar to ACN's ability to move their summon to a specific location, and similar to THM/ACN/BLM/SMN Limit Break). Move the targeting reticle over the glowy spot and left click. This should make you throw the key item at the glowy spot.

This is actually covered in the in-game help menus, so if you have that turned off, you might want to turn it on again.
