(1) Hamlets are not directly tied to the GC. Anyone can participate in any Hamlet so long as they're in a full party and everyone is L45+. There are no quest pre-reqs.
(2) No. Gear, anima (so much anima), and Beastman Seals for the Relic quest are the rewards. (Well, you also get DM as a consolation prize sometimes.)
(3) Depending on your abilities and gear, it can certainly be done at 45. The content isn't particularly hard if you just follow the accepted strategy. I've heard you want your Gathering to be above 90 if you decide to bring an L45 DoL, so you won't be able to snag an extra couple hundred points from using a broken low-level weapon.
(4) Yes. There is an easy, successful strategy that's used. Search for "Hamlet Guide" on the forums and you should find a couple guides. I'll summarize the party roles though. You'll want two WHMs, a tank, 3 DoH, and two DoLs. The DoHs should focus on keeping Defense Up and Attack Up active as much as possible, and the synth you must do has no benefit from quality, so equip abilities that guarantee success or give fixed progress. The DoLs will coordinate in keeping Attack Down on the enemies by gathering red potions, but can also use blue potions to cause a poison effect that will speed up the run. The WHMs keep everyone alive, especially the NPC archers. Use Regen! The tank's job is to kill mobs that spawn at certain times for bonus points, assisted by WHMs when needed, and then to kite the enemy leader around the area to delay the end of the fight for a particular bonus enemy to spawn.
