To answer the original question though, the "once-in-a-lifetime" content is anything dealing explicitly with the 7th Umbral Era. This is mostly the Grand Company quest lines we have currently, a few side quests, that giant red thing in the sky and possibly some of the Primals. There is content that will no longer be available once 2.0 hits.
