Only the part where the handmaiden—and her descendent—were Roes instead of Hrothgar would have to be introduced. In retrospect, it's really not that damaging.

It's still weird, though, that we've got this ragtag army of ex-soldiers and laymen turned resistance fighters of all races and creeds, and opposite them we have an entire Imperial Legion of similar make to the resistance—with some particular standouts like Daguza and his band of beastmen—but not a single one of them is a female Hrothgar.
Yes, I don't think the plot of the story would have played out differently with the betrayals, but all these racially diverse females all over the place should have been female Hrothgar. It hurts the story because the Hrothgar Queen become villain isn't racially Hrothgar. It has essentially stopped being a story about Hrothgar and their civilization and instead become a generic war story.

The faux pas is an equivalent if a white actor played Eric Killmonger in 2018's Black Panther. Yeah the plot would be unaffected but they've essentially stolen the villain's identity by casting the wrong type of person.

A roe becoming the Hrothgar Queen villain was a terrible choice for the story and ultimately undermines the Hrothgar specific content. At the very least they could have made 3 or 4 female Hrothgar npcs to fill in for Blade squad, the Queen/Misija, and females standing around Gangos and Southern Front base.

And when the players whined that they can see female Hrothgar and should be able to play them, SE could have stuck to their guns and said those female Hrothgar were essential to story.

But it's too late to retcon it, so the damage has been done.

The fact that the Queen is a Roegadyn instead of a Hrothgar is... Literally the point though. Even if we had female Hroths, Misija and the Queen still wouldn't be Hroths.

That Roe was a temporary queen only made queen to be a sacrifice. She had no real rule. She was just a handmaiden or something that they made become queen for 15 minutes to become a primal and then killed her off when she wasn't needed anymore. I don't really see the big deal there. If the original long term queen had been a Roe then yeah I'd get the complaint, but the sacrificial queen? I don't have a problem with that .

I would have preferred we just got one race. I would have been happier with both genders of Hrothgar OR both genders of Viera, either one, than the split we got. But I don't feel like this ruins Bozja's story or anything.

Do we know the gender split among Hrothgar?

I vaguely recall it being about 1 in 10 Miqote being male, but I can't imagine the difference being quite so huge among the Hrothgar, it just doesn't make a great deal of biological sense.
