It has to be Zenos. He just fits the role too well. We could argue all day about who the "real" villain of FFXIV is but we all know Zenos has more star power.
If we're going off the current story, Spiritus has been more about summoning the more morally grey villains, maybe because of the instability of the crystal cores of the world, and not trying to bring in characters that will make things worse. We saw that with characters like Leo and Gabranth being added in, and the Turks being added next month. We don't have any of the Act 3 story yet to know how that will effect things, unless you can read the Japanese, but unless they've fixed that, I would say that the most likely choice is someone who is less overtely power hungry or evil and more someone in that middle ground.
So to that end I would say Gaius is the most likely choice. That being said, since Opera Omnia was originally meant to be a spin off of NT, that game introduced Zenos, which makes him the more likely choice. Though considering what a BORING and one-note villain he is, I really hope they don't go with him for Opera Omnia.
Also if they do add one of the ascians, it's fair to say that they could show up without being summoned explicitly since they can travel between the shards, it's not much of a stretch to say that they could just come to this world on their own. That is, afterall, how Gilgamesh came to that world.

It would more than likely be zenos since he’s the xiv antagonist in Dissidia NT. I could see them releasing Shadowbringers version of y’shtola as an alternate character in the same chapter too.
