Once you've unlocked access, you can queue from anywhere using the Duty Finder. All the "Entry Point" really does is open up the Duty Finder with that particular instance selected.
So, nah, you can queue from anywhere, once queued, you don't have to stand around, go do anything you like wherever you like.
Yeah, once you queued up you can do "whatever" you want and go whereever you like.
However, you cannot Change the party setup - Inviting or dismissing party members will abandon the queue and you have to re-queue. You can progress in other quests but cannot attend solo duties. If you are in a cutscene when the duty finder pops, you have to finish / abandon the cutscene in order to accept the invite (you have 40 seconds to do so) or you will be withdrawn and have to queue again.

I feel that it’s important to add that you may also change your class whilst in a queue, but you will have to change back to the class you entered the queue with to accept once a match has been found
Also most cutscenes will be reset to the beginning if a queue pops whilst you’re in it so don’t worry about skipping cutscenes to accept a queue
