You need to have cleared Copperbell Mines to even unlock POTD, so that requires burning MSQ to level 17. Previous poster is correct in that you should burn MSQ as much as possible, then POTD between any notable gaps in the next quest in the MSQ chain, since you have to do MSQ anyways to unlock further content.
