It is very clear that Anima Relic will begin with a quest, just like the relic stages before it. And while it will no doubt have trials of some degree, I have no idea why anyone would think you need AS4 for it.

The only thing confirmed as 'Needing to start with' is needing the Zeta to skip the first phase of one Anima relic; Which of course is an optional perk.
