Rune Knight is just a variant of Mystic Knight, a.k.a. Sword Mage. If they do add it it'll probably be a tank class.
... but good news, chances of Samurai being the next tank class are pretty high! It was a tossup between Dark Knight and Samurai for being in 3.0, and they decided on Dark Knight because they didn't want to put in an Eastern-culture heavy class so soon after NIN. We already have Doma (Eorzea!Japan), so it's just a matter of waiting for Samurai refugees to arrive or going there ourselves...
If we're going to talk classes we'd like, again, I don't particularly care, but...
Red Mage would be cool, being the only class from the original Final Fantasy not yet represented. That said, I can't see how it would work in XIV since it's intended to be a mix of White and Black Mage, both of which are already in-game and eclipse Red Mage in terms of sheer power on either axis. It's a valuable class in old-school RPGs, but I can't see how they would implement it with XIV's strict combat roles.
Geomancer would also be neat as a caster DPS. Be interesting to see how they implement it.
If not Samurai, then I'd like Viking to be the next tank job. It's not the most iconic, but it's still pretty cool and not hard to design IMO. Very similar to WAR though.
Blue Mage, obviously, is one of my most desired options. Being iconic to Final Fantasy fans as well as one of the few classes the franchise can truly call its own, it's something they almost have to implement in the future.
For healing jobs, something like Druid or Shaman would be neat, but given CNJ / WHM already have elements of those classes I don't see how that would happen. Not particularly attached to those classes, either.
Beyond that, I'd like to see what the XIV team can cook up.
