The new jobs have only 1 or 2 requirements:
Level 50
Have finished the 2.55 mainstory.
There are no class requirements, but it's recommended you have the two classes leveled for cross-class purposes for the new 3 Jobs.
I completed main storyline, but where are you guys getting that doing that is needed for getting access to Ishgard?
Everything I've heard has seemed to say you could just buy the combo ARR+Heavensward combo pack if have never touched FFXIV and are good to just go start on one of the new jobs. I mean, honestly I think it would be kind of cool if there was some questing involved to "unlock" the jobs. However, so far, maybe/maybe not need a job at lvl15 to unlock the job (just like every other job), besides that, I see no evidence. You could be a dark knight running around with a sapling icon on your name... I'm prepared to see them!

Additionally, the guilds for the three new jobs will all reside inside Ishgard, therefore you’ll first have to come to Ishgard if you would like to become one of these new jobs.

First you will need to finish the main story which will also require you having a level 50 job. You cannot enter Ishgard until then and Ishgard is where the new jobs are started.

In order to visit Ishgard, you’ll first have to complete the main scenario quest “Before the Dawn,” which will be implemented in Patch 2.55 at the end of this month. For those of you thinking about returning at the start of Heavensward, we recommend coming back a bit earlier to finish off the main scenario quests. With the main scenario completed, you can install Heavensward and immediately accept the first quest of the expansion that will lead you to Ishgard.
Furthermore, those of you who have pre-ordered the all-in-one pack, FFXIV: Online, will receive the pre-order in-game bonuses but it will not come with Early Access. If you'd like to participate in Early Access, please purchase A Realm Reborn, finish the main scenario, and then pre-order Heavensward.

I have another question if you have one character on your account who has met these requirements can you create a new character who starts off as one of the new classes
Highly doubtful.
This game is not alt-friendly in any way.
EDIT: You can't even start as Rogue on an alt, so yeah...

Nope. The new character starts with all jobs at level 1 and no Main story completed. As you need access to ishgard on each character (which requires 2.55 to be completed) a brand new character cannot start as a new job regardless of whether you have another character that has unlocked them.
