I'd have to double-check the story scenes, but I thought he replaced the inquisitor before the inquisitor got there, and just told everyone that he was the guy they were expecting. No glamour required, because he just lied his butt off."Guillaume" is a bit weird, since it seems he was already using a Glamour to appear as the real Inquisitor Guillaume. But when you defeat him and dispell the dragon Glamour, he goes back to looking like the man whose identity he stole. Not sure what that means, other than the strong possibility someone made a mistake somewhere.

I got the impression that some knew him by face, though that may have been in the context of having seen the impostor earlier-but-after-he-started. In any case, he seemed very high profile. It's kind of absurd to go in assuming that there wouldn't be anyone who could say, "Nope, your face is wrong!" Not when you have access to appearance changing magics.
There was also the corpse of the real inquisitor. I'm kicking myself for not checking closer, but I thought he looked the same.
