By the way, I don't know if any of you played World of Warcraft just after the release of Wrath of the Lich King (EU side) but for a while it became impossible to enter instances as too many were in play, due to the way the lockout system worked - they eventually sorted it by adding new servers and general changes to the lockout system (as it had not had one for standard five man dungeons before then! Oh Ramparts, how I loathe ye), but restrictions were placed upon group sizes until they got it sorted out (ie, no solo instancing)
Same deal here I'd assume, they are not limiting it as they honestly don't want people to try and solo it, or do it with a member or two less, but just to reduce server load by not opening up extra instances. They did say that after all, and it makes perfect sense - though hopefully in the future they can let folk do so if they wish.

It will be even worse if there is not a Reservation System in place. Imagine you've been waiting 59 minutes for the next Instance and someone else just walks up and gets lucky and gets in ahead of you.

Obviously, I can't say for sure what he meant. But I find it very difficult to believe that only one group can be in the dungeon at one time. I mean, that would be so dumb it's just not realistic. As much as they've been hyping up these dungeons, even going so far as to change the focus of the game away from leves and toward dungeons, there's really no way they're going to limit them to one party at a time. They know we're going to make a mad rush for those dungeons as soon as they're released. Forcing us to wait in week-long lines after release would be disastrously stupid.
I think he's suggesting that there is a physical limit to the number of parties who can be running the dungeons at one time. That's just practical reality, since the dungeons will be hosted on instanced servers, and the servers can't hold an unlimited amount of data. Eventually, the servers are going to fill up, and more players won't be able to get in without the dungeons running really, really slowly for everyone. That would also be bad, so they have to put a limit on the number of parties that can run them at one time.
As Yoshi-P says, that's another reason why they require parties of at least 4 or 8. If people are allowed to solo them, there will be 4 times as many r30 dungeons being run at one time, and there will be 8 times as many r50 dungeons being run at one time. So the servers would fill up much more quickly.
I also suspect that the number of players trying to run dungeons -- and as a result, any potential wait times -- will be worst right after the dungeons are released. But maybe that will be mitigated somewhat by the precursor quests that you're required to complete before you can enter the dungeons.
