The final nodes are a different type than everything that came before. Every slot has a question mark bag on it, but you don't what is behind them, or which have nothing behind them at all. Toil changes it to something akin to what you are used to dealing with.

Also, each slot of an unspoiled node has a "Health Bar" you need to deplete by gathering a few times before you can start getting the actual item. Toil of the mountaineer automatically depletes all of these health bars and lets you start getting items right from the start.
That's what the level 50 special nodes look like. You need to smack a spot to remove the 'health bar' before you can gather an actual item. Unless your stats are trash, it should only take one hit. That hit does remove from the node's 'durability' though so Toil gives you one extra swing to play with (particularly useful if you want to use the ability that guarantees HQ after a successful chain).
Items are always in the same spot (and empty spots always empty) so you can just memorize them/take notes/find a spreadsheet. Since toil removes all those 'health bars' at once you it also means you don't have to remember/reference said notes/spreadsheet.
The 50 nodes that the mechanic applies to only show up at certain times of the day and have 6 swings worth of durability before going away, thus GP gear is suddenly useful (after being borderline pointless while leveling) as you can blow it all on the node and then let it regen while you wait for the next node to pop up (likely elsewhere in the world).
I see I see, I haven't encounter this yet, but will it always say out of reach even if you uncover them the first time?
Yes, every node you visit of the one star materials will have that 'out of reach' screen to deal with. You can't access them until level 46 since they're only in that final 46-50 group.
