No, you can transfer while it is closed. I did that when I moved servers, I did it at like 7pmCST and got it through to a locked server in less than 10 minutes. Start the process at the Mog Station, if your worried about it taking a long time, then complete it before bed incase it does take a bit longer than it should.

You could also buy up stuff that's cheap on your world and sell high on new world. But you need a contact on the new world to sort out prices obviously.
I did that when I moved my legacy char over to my new server, made a couple million over what I had :)
