Keep up to date on the main story quests (flaming quest icon). And every 5 levels go back to your guildhall to pick up your next class quest. (If you add additional classes, you'll have class quests for each of them as well to keep up with.) Once you unlock your job, class quests are replaced with job quests, and you have to go wherever the job NPC is rather than the guildhall. (The initial quest to unlock a job is picked up at the main class's guild-hall, and you'll learn during that quest where to go for the additional job quests each 5 levels thereafter. In the case of Bard, the NPC you need to talk to is in South Shroud.)
The combination of main story quests and class/job quests will unlock the vast majority of the content.
A few things, though, are unlocked by side quests:
- The ability to dye your gear is unlocked by a level 15 side quest in Western Thanalan.
- The ability to convert used gear into materia is unlocked by a level 19 side quest in Central Thanalan. (The main story quests will take you to the right area. When you meet the character who tells you all about materia, take the sidequest from the other NPC standing next to him.)
- The ability to use mounts is unlocked by a side quest at your Grand Company's headquarters once you join a Grand Company at around level 20. (Again, the main quest will have you there when that quest first becomes available. Take the chocobo sidequest at your GC headquarters as soon as your membership in that GC is official.)
- The ability to have your chocobo fight alongside you is unlocked by a level 30 quest in the South Shroud.
The other thing that could be missed is guildhests. They first become available at level 10, and a couple more open up each five levels, but while the main storyline initially at level 10 tells you about them, there's no requirement to do them right away and many people never bother. Then you get to endgame and come across a quest that requires you to do the last of them, but they have to be done in order, meaning you have to start with that initial level 10 guildhest and work your way up through all the rest. So if you prefer doing stuff when you're at the right level for it, add guildhests along with your class/job quests as something to do when you hit each fifth level.
